HIV Vaccine Candidate Imbokodo Advances into Phase 2 Study

Scientists announced that a trial drug against the AIDS vaccine has yielded encouraging results and has advanced to the next phase of the pre-approval trial process

While these results are not ideal, they "represent an important milestone", Harvard professor, Dan Barouch, said.

In humans, the vaccine was safe and produced antibody responses and T-cell responses, and in monkeys, the vaccine not only produced similar immune responses, but protected against a type of simian-human immunodeficiency virus (SHIV), reported Dan H. Barouch, MD, of Beth Israel Deaconess Medical Center in Boston, and colleagues.

Barouch reminded that this is the "fifth HIV vaccine concept" that has been tested in the four decades of the advent of HIV. The achievement was considered to be too small for the drug to go ahead and come into the market as a HIV vaccine.

Based on the results from this phase 1/2a clinical trial that involved almost 400 healthy adults, a phase 2b trial has been initiated in southern Africa to determine the safety and efficacy of the HIV-1 vaccine candidate in 2,600 women at risk for acquiring HIV.

This study was supported by Janssen Vaccines & Prevention BV and the NIH, the Ragon Institute of MGH, MIT and Harvard, and a cooperative agreement between the Henry M Jackson Foundation for the Advancement of Military Medicine and the US Department of Defense.

The mixture of HIV strains in the "mosaic" vaccine is delivered using a nonreplicating common-cold virus. This includes testing the drug on a much larger group of people.

The randomized, double-blind, placebo-controlled study showed that the vaccine evoked an anti-HIV immune system response in all 393 of the patients who received the treatment.

Now, scientists are making significant strides toward an alternative solution: HIV vaccines.

Scientists announced Saturday that they had already piloted an experimental HIV vaccine, which triggered an immune reaction in lab animals and successfully immunized monkeys from the infection.

A parallel study in rhesus monkeys was also performed so that immunogenicity and protective efficacy of the various vaccines could be assessed and the optimal vaccine regimen could be realised to transfer to further clinical studies.

However, scientists in the USA have created a vaccine that has managed to get to the first step, with nearly 400 healthy uninfected adults mounting an immune response to the vaccine.

With an estimated 1.8 million new cases of HIV every year, a safe and effective vaccine would be an incredible tool in stopping the pandemic. The participants were treated in clinics in east Africa, South Africa, Thailand, and the US, and either received a placebo or one of seven vaccine combinations.

Participants in the research were selected from 12 clinics in East Africa, South Africa, Thailand, and the USA.
