Scientists Discover New Class of Antibiotic to Fight Drug-Resistant Superbugs

Antibiotics family discovered in soil could be the answer to antibiotic resistance

In order to hasten the process of culturing soil bacteria in a lab, the researchers used high-speed computer processing to "screen" their soil sampled for this calcium use. That's where numerous most widely used antibiotics in medicine come from.

The identification of this new antibiotic is similar to that of penicillin, the world's first antibiotic drug first discovered in mold bacteria by Scottish scientist Alexander Fleming in 1928.

But an even more singular event would be the discovery of a new class of antibiotics that doesn't prompt the development of resistant strains of bacteria.

After infecting rats with MRSA, a bacteria that's particularly resistant to most antibiotic treatments, the researchers treated the rodents with malacidins, and the compound eliminated the infection in the animals' skin wounds.

The researchers are now working to improve the drug's effectiveness in the hope that it can be developed into a real treatment for people.

The result has been called a "slow catastrophe": the Centers for Disease Control and Prevention estimate that each year, at least 23,000 people now die as a direct result of bacterial infections that have become resistant to existing medicines. Prior to that there had been a 30-year drought, prompting dire warnings of a "post-antibiotic apocalypse" if bacteria continue to adapt to withstand essential drugs.

The team of researchers led by Sean F. Brady, a chemist and associate professor, and his colleagues at Rockefeller University in NY looked at the bacterial DNA from 2,000 soil samples that were taken from different parts of United States.

Brady's team was looking specifically for distant relatives of daptomycin, which uses calcium to bust up, break down and generally disrupt the cell walls of target bacteria. "It is a long, arduous road from the initial discovery of an antibiotic to a clinically used entity".

Leading researcher of the discovery, Sean Brady, PhD, who is an associate professor and chemist at the Rockefeller University said in a statement that, "We're missing most of the molecules that would have come from that extraordinarily productive platform". When they noticed a number of samples contained malacidins, they made a decision to dig a bit deeper into the compounds.

Brady added that, "We're taking DNA directly out of soil samples". By narrowing their search for the DNA signature of calcium dependence, they were able to find a needle in a haystack - and find a promising compound. The team next cloned the relevant genes and rearranged them to insert into a host organism Streptomyces. "We'll see where it goes from here", he said.

Brady says there are at least 10,000 bacteria under each footprint of soil, majority still unidentified.
